Objective:(1) The aim of the study was to examine T-helper(Th)17cells and interleukin(IL)-17expression in peripheral blood of patients with the primary immune thrombocytopenia (ITP).(2) The aim of the study was to examine Th22cells and IL-22, tumor necrosis factor-α (TNF-α) expression in peripheral blood of patients with ITP.Method:(1) The proportion of Th17cells from48cases of patients with ITP and the healthy control was detected by flow cytometry (FCM), and the cytokines level of IL-17was evaluated by enzyme-linked immunosorbent assay (ELISA).(2) The proportion of Th22cells from48cases of patients with ITP and the healthy control was detected by FCM, and the cytokines level of IL-22and TNF-α was evaluated by ELISA.Results:(1) Compared with the healthy control group, the percentage of Th17cells was increased in ITP patients (1.40±1.28%,0.70±0.45%,p<0.05), it was significantly lower in the glucocorticoids hormone treated group than untreated group (0.60±0.52%,1.55±1.11%, p <0.05). The level of IL-17, expressed by Th17cells in ITP patients was increased compared with the healthy control group (19.62±5.19pg/ml,18.50±3.58pg/ml,p<0.05), it was, however, lower in the glucocorticoids hormone treated group than untreated group, but there were no statistically significant difference between the glucocorticoids treated and untreated group(p>0.05).(2) Compared with the healthy control group, the percentage of Th22cells was increased in the ITP patients (0.55±0.30%,0.26±0.22%,p<0.05), it was significantly lower in the glucocorticoids hormone treated group than untreated group (0.43±0.16%,0.60±0.32%, p <0.05). The level of IL-22, expressed by Th22cells in the ITP patients was increased compared with the healthy control group (112.16±22.66pg/ml,92.78±41.01pg/ml,p<0.05), it was, however, lower in the glucocorticoids hormone treated group than untreated group, but there were no statistically significant difference between the glucocorticoids treated and untreated group (p>0.05). The level of TNF-α, was increased in the ITP patients compared with the healthy control group (15.73±4.46pg/ml,11.89±1.87pg/ml,p<0.05), it was, however, lower in the glucocorticoids hormone treated group than untreated group, but there were no statistically significant difference between the glucocorticoids treated and untreated group (p>0.05).Conclusions:(1) Our study demonstrated that Th17may play a role in the pathogenesis of ITP, and the glucocorticoids hormone may probably play a therapeutic role by regulation effects on the Th17cells.(2) Our study demonstrated that Th22may play a role in the pathogenesis of ITP, and the glucocorticoids hormone may probably play a therapeutic role by regulation effects on the Th22cells.
