Objective:To investigate the effects of celastrol on the apoptosis and cellcycle arrest of C6glioma cells and its mechanism.Methods:C6glioma cells were cultured in vitro. MTT was used to assaycellular proliferation. Fluorescence microscope and transmission electronmicroscope were used to evaluate the morphological alterations of C6gliomacells after being incubated with2.56μmol/L celastrol. Flow cytometry wasused to analyze the alterations of apoptosis rate and cell cycle.Results:The inhibitory effects of celastrol on C6glioma cells weredependent on celastrol concentration and incubation time. Typicalmorphological alterations of apoptosis were observed under photo-microscope,fluorescence microscope and transmission electron microscope respectively.Flow cytometry showed that the apoptosis rate increased from1.32%incontrol group to19.34%in celastrol group, and cellular quantity decreasedfrom76.42%to54.52%at G0/G1phase but increased from9.29%to30.50%at G2/M. Western blotting analysis showed that the expression ofapoptosis-related proteins Bcl-2and XIAP were down-regulated, but Bax,Caspase-3and cleaved PARP were up-regulated. Moreover, the expression ofcell cycle regulator p21, p2and cyclin B1were increased, but cdk2wasdecreased.Conclusions:Celastrol could induce C6glioma cell apoptosis and cellcycle arrest via influencing the expression of apoptosis related proteins and cell cycle regulators.
