Objective:Through the study of alkB gene and its protein in gastriccarcinoma and away from the tumor, normal mucosa tissue expression, inorder to clear alkB gene expression in gastric cancer and precancerous lesionsand relationship. Through folic acid on gastric cancer cell lines MKN-45afterits alkB gene and protein expression changes with the preliminary discussionof folic acid in the treatment of gastric cancer and the mechanism of alkBgene and its expression product and the occurrence of gastric carcinomacorrelation.Methods: The first part: select60patients with operation or biopsy ofgastric cancer patients with gastric cancer, cancer, cancer adjacent to andaway from the normal mucosal tissue, using RT-PCR and Western Blot weredetected in the tissues of alkB gene and protein expression, analysis of itsgene and protein expression in gastric cancer and correlation; part second:selection of protein expression gastric carcinoma cell line MKN-45, folic acidand RT-PCR and MTT method, the detection of alkB gene and proteinexpression changes, in order to clear the regulation of alkB gene methylationlevels relationship with gastric carcinoma. Results: AlkB gene in breast cancer from normal mucosa tissueexpression is strong, in the adjacent mucosa tissue expression decreased,while the expression in gastric cancer tissue and more weak;0.2mg/L,0.4mg/L concentration of folic acid could obviously inhibit the proliferation ofgastric cancer cells with different concentration of folic acid interventionactivity; gastric MKN-45cells after72h,0.2mg/L,0.4mg/L folate groupalkB gene mRNA expression was significantly higher than that of controlcells (P <0.05).Conclusions:Gastric carcinoma alkB gene product repair DNA andaberrant methylation of RNA damage ability may decline, down-regulation ofalkB gene expression may be involved in gastric cancer tumor suppressorgene methylation of abnormal high expression and silencing, namely thedown-regulation of alkB gene expression may be involved in gastriccarcinogenesis mechanism; folic acid can inhibit gastric carcinoma MKN-45cells proliferation activities, at the same time upregulate the expression ofAlkB geneļ¼Œof which0.2mg/L,0.4mg/L concentration of folic acidinhibitory effect is the most obvious. Thus, folic acid intervention couldeffectively inhibit the growth of gastric cancer cells.
