Object iveAt first the paper observes the predicaments symptoms and the effect of high fat diet to the body by the high fat diet production model. Then we make the type2diabetes models by using high fat diet combined with small dose of intraperitoneal injection of STZ and study the substance metabolism of the type2diabetes models.At last, we observe the effect of recipe for supplementing Qi and Buzhong Yiqi Decoction by the differences among groups.Methods(1)Model oneMale rats randomly divided into four groups. Normal group (feeding ordinary diet); model group (feeding high fat diet); RSA1, feeding RSAl (S:A=2:1)by gastrogavage; RSA2, feeding RSA2(S:A=1:2)by gastrogavage. NO3-, ONOO-, MDA, glucose and amylase activity in rat faeces were determined by colorimetric method, N-glycan patterns of amylase were determined with lectin (PSA and PNA) affinity precipitation method, and faeces amylase isoenzyme profile were separated by electrophoresis on Polyacrylamide gels electrophoresis (PAGE).(2)Model two61male rats are randomly selected from10as normal control group; the rest of the SD rats were used for models, the models (after72hours of fasting blood glucose values:above16.7mol/L) are randomly divided into model groups, treatment one.treatment two. Normal group is given normal diet; the others are fed high fat diet. The treatment I is fed with Buzhongyiqi decoction by intraperitoneal injection; the treatment Ⅱ is fed with Buzhongyiqi decoction which is treated by hydrochloric acid by intraperitoneal injection. The model group and normal group are fed with physiological saline by intraperitoneal injection. Then we measure the urine pyruvate, lactate, creatinine, free fatty acid, urea nitrogen, urinary metabolites of N03-, ONOO-, lipid peroxides MDA, GLU and make a statistical analysis.Results(1)Model oneHigh fat diet rats showed the symptoms of polyuria, polydipsia, polyphagia and weight loss, these abnormal signs were improved after treatment with RSA. In high fat diet rats, amylase activity, NO3-, P-Amy, S-Amy in faeces were decreased, while PSA, PNA binding rates of amylase, glucose,ONOO-in faeces was increased, after treatment with RSA, all of which were improved, there is the difference between RSAl action and RSA2action.(2)Model twoThe model rats gradually emerge polyphagia, polyuria, weight loss and other symptoms. The symptoms of the treatment groups are significantly improved.Conclusion(1)Model oneSupplementing Qi and activating blood can improve N-glycan deficiency in faeces amylase and up-regulate faeces amylase activity, and increase N03-, also decrease glucose, ONOO-in faeces, thus can regulate the digestion/absorption and glucose metabolism, can reduce oxygen free radical-induced intestinal damage. This study also showed the different impacts of two different RSA.(2)Model twoBuzhong Yiqi Decoction can improve the symptoms of the type2diabetes rats. The Buzhong Yiqi Decoction can improve the metabolic disturbance the type2diabetes rats by improving the function of the spleen. In other words; it has the function of improving insulin resistance syndrome.
