ObjectiveWe research the radioprotection of lentinan respectively on the organic, cellular and moleculary level, meanwhile investigate the mechanism of radioprotection. Looking towards to provide some experimental base for the research of polysaccharose.Methods1. We make the model of radiation-damage mouse by ionic accelerator, using0.4Gy as absorbed dose.2. We observe the psychosis, record the weight and the state of skin, calculate the organ coefficient, make pathological section of every viscera, in order to analysis the integral radioprotection of lentinan.3. We detect the activity of SOD, GSH-Px and the contents of MDA using ELISIA kit, observe hepatic ultrastructure using transmission electron microscope, in order to analysis the radioprotection of lentinan for hepatic cells.4. We detect the proliferation activity of splenic lymphocytes using MTT assay, detect the secretory volume of IL-2of splenic lymphocytes using ELISIA kit, in order to analysis the radioprotection of lentinan for splenic cells.5. We review the quantitative and qualitative expression of proteins which expressed on the membrane of splenic cells using one-dimensional SDS-PAGE and MALDI-TOF/TOF, in order to analysis the radioprotection of lentinan for biomacromolecule.Results1. LNT could protect multi-viscera injures:LNT could avoid slip of hair obviously; regulate the fluctuation of weight by preventing the weight increased rapidly in the earlier stage of radiation and the weight loss in the late stage of radiation; maintain the normal level of organ/body weight ratio(including spleen, thymus, liver) and microstructure(including spleen, liver, lung).2. LNT could protect cellular injures:LNT could enhance the activities of antioxidant enzymes; induce the synthesis of oxidation products,such as MDA; protect the secretory function and ultrasturctural of plasma. LNT could stimulate the activity of lymphocytic proliferation; increase the secretion of IL-2from splenic lymphocytes; protect the ultrasturctural of plasma.3. LNT could protect biomacromolecule injures:LNT could regulate the expression of proteins which relatived to protein and lipid metabolism, cytoskeleton, transfer of protein and ion.ConclusionLNT could provide certain radioprotection on the organ, cells and biomacromolecule. These protection may all interrelated with the action of resistance to free-radical and the action of regulate the expression of proteins.
