Objective:To perform a meta-analysis to evaluate the risk of leucopenia/neutropenia associated with bevacizumab.Methods:Medline, Embase, Cochrane Library and Chinese Bio-Medicine Database were searched to identify relevant studies. Eligible studies included randomized controlled trials for comparing the combination of bevacizumab and chemotherapy with chemotherapy alone for cancer patients. Patients were enrolled according to pre-specified eligibility criteria for each trial. Data were extracted by two reviewers using the designed extraction form. Review Manager5.1software was employed for the meta-analysis. A fixed-effect model or random-effect model based on the heterogeneity test were used to analyze data.Results:Eighteen clinical reports were enrolled in the study which included a total of11777patients. The results showed that the relative risk(RR) of leukopenia in the group of bevacizumab combination chemotherapy and control group is1.19,95%confidence interval [0.93to1.52]; test for overall effect Z=1.37, P=0.17, P>0.05, the difference was not statistically significant. Compared with non-bevacizumab therapy, the group of bevacizumab and chemotherapy has a slightly increased risk of neutropenia (RR=1.07,95%confidence interval [1.02to1.12], P<0.05) and increased risk of febrile neutropenia (RR=1.59,95%confidence interval,[1.33to1.89], P<0.00001).Conclusions:Treatment with bevacizumab is associated with a slightly increased risk of neutropenia and a increased risk of febrile neutropenia with chemotherapy in cancer patients. Early detection and prevention neutropenia especially febrile neutropenia have important clinical significance for patients accepting of bevacizumab and chemotherapy combined treatment.
