Objective:To investigate the neuroprotective effects of MaFGF on hippocampal neurons exposed to cisplatin (DDP) in vitro.Methods:Hippocampal neurons from neonatal SD rats were primarily cultured for seven days and identified by immunocytochemistry of neurofilament heavy polypeptide (NF-H). These primary cells were inoculated into6-well plates,24-well plates and96well plates.①7-days later, different concentrations of DDP were added, and MTT assay was performed to test the cell viability after24h.②The injury model was induced by DDP, and different concentrations of MaFGF would be added simultaneously. Finally, the protective effects were observed.③The cells were divided into three groups randomly:Control, DDP and MaFGF+DDP group. Control groups were incubated normally; DDP groups were incubated with medium containing25μg·ml-1DDP; MaFGF+DDP group were incubated with medium containing5.2μg/L MaFGF, all of them were curtured another24hours. And then, the preventive roles of MaFGF in the injured hippocampal neurons were observed by using inverted phase contrast microscope and some biochemical and enzymological indexes.Result:(1) The cell vitality reduced when increased the concentrations of DDP, which indicated that DDP inhibited the vigor of neurons significantly. The hippocampal nerve cells were swelling with vacuoles existing in cytoplasm, even some cells were sheded and suspended in culture medium.The neurites atrophied and reduced.(2)The survival of hippocampal neurons added with DDP increased obviously with MaFGF between5.2to10.4μg·L-1. The cells were well protected by MaFGF. We could see that the swelling of cells and the loss of neurites were Significantly reduced.(3) There were statistical differences of biochemistry and zymology between MaFGF+DDP group and DDP group (P<0.01), the activity of SOD and GSH-Px of MaFGF+DDP group increased while the lever of ON and MAD decreased.Conclusion:(1) Cisplatin could cause the injury of hippocampal neurons in vitro, which was worse when increasing the concentrations of DDP.(2)MaFGF might play a significant neuronal protection role in hippocampal neurons injured by DDP in some degree.(3)The preventive effects of MaFGF on DDP neurotoxicity were related to antagonize free radicals directly or indirectly, enhance the activity of antioxidant enzymes which could scaven free radicals, and protect antioxidant enzymes.
