It has already entered the aging society since1999in China. Growth of the elderly population, resulting in a series of elderly patients such as ischemic cerebral vascular disease is increasing year by year. This type of morbidity, because of the high rate of disability, mortality and relapse, as a serious danger to human life and health, has become one of the three major diseases in our life, bringing heavy burden to family and sociaty. Cerebral micro vascular plays an important role in maintaining stability in the neurons.During ischemic injury, belonging to ischemia time extension, the number of micro-vascular is gradually increased. Research indicates that the micro vascular ischemic Penumbra density increases survival rates in patients with high regeneration of ischemic vascular and cerebrovascular disease closely related to development outcomes. Studies on vascular regeneration after acute brain injury has also become a focus.Granulocyte Colony Stimulating Factor, G-CSF was first found by Metcalf as a vivo polypeptide chain founded cell growth factor in1980. Its function mainly in the area of specific regulation of myeloid differentiation and proliferation, increased granulocyte function and on defense stress function. G-CSF-R present in neurons and glial cells, which tips that G-CSF plays an impotant role in injury and physiological of the nervous system. Nowadays, it is attracting widespread concern with the field of neuroprotection. Possible mechanisms of CSF mainly contain anti-apoptosis, proliferation and migration of endogenous stem cells differentiation, anti-inflammatory, and block the glutamate excitotoxicity, as well as promoting angiogenesis and so on. Angiogenesis is a process of budding in the original blood vessels on the basis of the formation of new capillaries, which core is splitting the proliferation of vascular endothelial cells. Expression of Survivin regulation of apoptosis factors such as Caspase-3, is an important anti-apoptotic factor.It is found that Survivin express strongly on the vascular smooth muscle, stereo tube endothelial cells in vitro and in vivo new capillaries recently.Inhibition of Survivin expression inhibits angiogenesis, forced expression of vascular endothelium and tiped in the angiogenesis plays an important role in the process. Egr-1features mainly involved in cellular signal transduction and regulation of expression of multiple genes, including growth factors, signal transduction genes, cancer genes and gene transcription factor gene, as well as playing an important role on growth, development, differentiation and cell injury repair. It is discussed that the effect in promoting angiogenesis factor and promoting angiogenesis in this artical.It played a catalytic function that the expression and secretion of these factors of Survivin needs involvement of the transcription factor Egr-1, while survivin is in effect of a range the expression and effection of Egr-1.Materials and MethodsIt has been randomly divided into3groups with SD rats aged60d in good health: the sham operation group, MCAO operation group and rhG-CSF treatment group.7d,14d,21d after the complete of model, each group filled with8samples, The parts which was choosed into the G-CSF treatment group was made subcutaneous injection with G-CSF50μg/(kg·d) only if it stay alive and accord with the standard of Zea Longa, totally quantity measure to7.5ml,lasting for5days. While the operation group wasinjected with7.5ml normal saline,the same is done with the sham operation group. To investigate the expression of Egr-1and Survivin uses the way of immunohistochemical. It expressed more in model group and obviously treated groups in positive cells.It is observed that Survivin main expressed in neurons, oligodendrocytes, astrocytes cells cytoplasm, as a small number it expressed in the cell nucleus, while significantly increased angiogenesis in particular active part-positive cells, Survivin models are more significantly in increased surgical group, at any time of the setting point in time,that useing the statistically significant (p<0.05).It reached peak in14d in reperfusion therapy group. Survivin expression became a marked increase in model group in the setting time point of7d,14days, that useing the statistically significant (p<0.05), with no significant differences in model group21d (p>0.05).The expression of Egr-1After focal ischemia-reperfusion the number of Egr-1-positive cells in the surgical group was scarce, and talking about dyeing. The number of expressing positive cells are more and dye stronger in model group and obviously treated groups. It is tha Egr-1expressed gradually from nucleus to cytoplasm. Egr-1are more significantly increased in surgical group than models group for each time point,that the differences are statistically significant (p<0.05). Egr-1reached peak in reperfusion treatment group at14d. Egr-1expression became a marked increase in model group in the setting time point of7d,14days, that useing the statistically significant (p<0.05), with no significant differences in model group21d (p>0.05).Analysis of relationship between Survivin and Egr-1model groupExpression of Survivin and Egr-1are related. r=0.314,P<0.05. Conclusion1.The expression of Survivin and Egr-1increased in rats with ischemia reperfusion injury. Expression are relevant beween each other, both of which participate in capillary in ischemic Penumbra.2.RhG-CSF increases expression of Survivin and Egr-1and accelerates capillaries, prompting rhG-CSF plays a protective effect of cerebral protection by roducing the number of capillaries in the damage area.
