| Chinese herbal is our traditional and peculiar medicine, and is the precious natural resources and precious wealth of the human. Fructus corni, one of the plant medicines, belongs to Cornaceae, Macrocarpium and also known as Arillus corni, Drugzao pi and so on. It is initially distributed in Henan, Shanxi province. Clinically, it has the effects on fulling liver and kidney, astringing essence and solidifying collapse. In the recent study, it has been found that it exhibits a number of biological activities, including reducing blood glucose, antiaging, immunological regulation and so on. And Celastrus angulatus, which is botanical pesticide, belongs to Celastraceae, Celastrus. The effective components have the antifeedant, toxic and anesthesia effects on the pests. A study has been worked out in order to make full use of Fructus corni and Celastrus angulatus, to further development other biological activities, and to find the chemical constituents of these plants.In this paper, the standard curve of Loganin was established by HPLC first. The contents of Loganin were used to screen the extracting dissolvent of Fructus corni. And the extracting dissolvent that was20%and95%ethanol was determined. Then the ethanol extracts from different polar parts of Fructus corni were separated and purified by HP20macroporous adsorption resin, and the optimal technological conditions of separation and purification were confirmed by the experiment. After this, with chloroform-methanol and petroleum ether-ethyl acetate as eluents, let the further processing extracts through the atmospheric pressure chromatography column. Then a systematic separation of the Fructus corni was worked out using preparative high-performance liquid chromatography. Through the NMR data analysis method, which including1D(1H,13C NMR and DEPT) and2D NMR (1H-1H COSY, HSQC and HMBC) techniques, the structures of eight compounds were identified. They were:loganin,7-dehydrologanin,7-a-morroniside and7-p-morroniside, ursolic acid, oleanolic acid, glucose, cornuside, cornuside â…¡. Two compounds of which, loganin,7-dehydrologanin, were completely analyzed and assigned using a combination of1D NMR and2D NMR. Then, the13C NMR fingerprint of Fructus corni was established for the first time, and a basic research was gained. Finally, the study of the triterpenoids from Celastrus angulatus was achieved, the structures of two known compounds and a new compound separating from it were identified. |
