AIM To construct the THP-1 derived foam cells formation and evaluate the effection on theexpression of ABCA1 and content of cholesterol respectively by the Ang-(1-7) and MDL. MDLis the inhibition of adenylate cyclase.METHODS THP-1-derived macrophages were treated with ox-LDL to construct the foam cellsfor the research. The foam cells samples were divided into 4 groups: Group A: control group:The foam cells without any treatment; Group B: foam cells treated with MDL (the inhibition ofadenylate cyclase); Group C: foam cells treated with Ang-(1-7); Group D: foam cells treatedwith both MDL and Ang-(1-7). At 24h after treatments, the expression of cAMP was measuredby Elisa. The mRNA and protein level of ABCA1 were measured by real time RT-PCR andwestern blot, respectively. The content of cholesterol was measured by high performance liquidchromatography.RESULTS The level of cAMP、ABCA1 mRNA and ABCA1 protein Group C were significantlyhigher than Group A(P<0.05)and the content of cholesterol was significantly lower than GroupA(P<0.05). On the opposite, the level of cAMP、ABCA1 mRNA and ABCA1 protein Group Bwere lower(P<0.05)and the content of cholesterol was significantly higher than Group A(P<0.05),The results of Group D were between Group C and Group A.CONCLUSION The Ang-(1-7) inhibited the formation of foam cells by promoting theexpression of ABCA1 and decreasing the content of cholesterol. MDL partly antagonize theAng-(1-7) by inhibiting the adenylate cyclase and decreasing the content of cAMP.
