Objective : To compare the separate and combined effects of active carbon absorption andepithelial curettage in corneal alkaliburned rabbits.Methods : 24 adult rabbits were randomly divided into 4 groups,6 rabbits each group.A: controlgroup.B: active carbon absorption group.C: epithelial curettage group.D: combined therapygroup.Then established the model of alkaliburned corneas in each right eye.After absorption andcurettage,same regular medications were applied to each group.At the projected timepoints,corneal fluorescein sodium staining and pathological changes under light and electronmicroscope were used to estimate the condition of corneal epithelial repair.Results :(1)The rate of corneal epithelial repair:After the model of alkaliburned corneas was made,each cornea was gradually healing as timewent by. At the timepoint of 30min,1,3,5,7d, There were statistical significance between groupA and other 3 groups,group D and the left groups(P<0.05). Differences between group B and Cwere not obvious(P>0.05).Of all the groups,combined therapy group was the best.(2) Observation of light microscope:On the 7th day after alkali burn,group A: large area of epithelium desquamation with infiltrationof a mass of inflammatory cells,and the left epithelium was distortion,and the collagen fiberswere loose and fractured.Epithelium of group B and C was mostly restored.There were someinflammatory cells,and some of the collagen fibers are disorderly ranged.Epithelium of group Dwas basically repaired,and there were few inflammatory cells in stromal layer.Collagen fiberswere more orderly than group B and group C.On the 14th day after alkali burn:Epithelium of group A was mostly repaired,but the thicknesswas uneven,and the collagen fibers were not well-distributed.The other 3 groups: Epithelium wasrepaired,the shape of epithium cells were regular.The collagen fibers of group D werewell-distribute compared with group B and group C.(3) Observation of Transmission electron microscope:On the 14th day after alkali burn:Group A:There were few connection between epitheliumcells,the colour of nucleus were light,the number of mitochondria and rough endoplasmic reticulum were small,the cristae of mitochondria was sparse. Collagen fibers in stromal layerwere loose and swelling.There were more mitochondria and rough endoplasmic in group B andgroup C,the colour of nucleus were still light.Group D: The number of connection betweenepithelium cells and mitochondria and rough endoplasmic was significantly increased,thechromatin of nucleus was rich, the cristae of mitochondria was intensive, the collagen fiberswere well-distribute compared with group B and group C.Conclusion :1. Distinctive differences are not seen between the separate therapy groups.2. Combined therapy group is superior to other ones.
