Objective: The purpose of this paper is to observe tensile stimulate on the invasion capacity and theexpression of matrix metalloproteinases-2(MMP-2) in the Tca8113 cells which achieved byFX-4000 flexible basement membrane loading system in vitro ,and investigate the effort of tensilestimulation in the invasion and metastasis of Tca8113 cells , thus for further to explore themechanics biological effort in the process of invasion and metastasis of tumor cells.Methods: Amplitude 2.0%, frequency 2.0Hz strain tensile were loaded on the Tca8113 cells, thestraining time is 4h,8h,12h. unstrained cells were used as a control. The expression of MMP-2were studied with Immunohistochemical techniques and Image software for quantitative analysis,The invasion capacity of Tca8113 cells was determined by Transwell in vitro, count the numbersof every vision ongue cancer cells which through the Matrigel glue and the results with x±s said.Results: The numbers of every vision tongue cancer cells which through the Matrigel glue of thestrained group was obviously increased than the unstrained group(P<0.05) and the numbersincreased as the straining time increased(P<0.05). the result of immunohistochemical showed thatthe expression of MMP-2 of the straining group is higher than the unstrained group((P<0.05) but itdidn’t change as the straining time changed(P>0.05).Conclusion: The tanswell experiment showed that stretching stimulation can promote the invasioncapacity of the Tca8113 cells and have the time correlation. as the same time ,the result ofimmunohistochemical showed that stretching stimulation can promote the expression of MMP-2of Tca8113 cells, but there was no time correlation. The result hints that Mechanical stimulationplays a certain role in the invasion and metastasis of Tca8113 cells. some protein other than MMP-2was influenced by the stretching may promote the invasion and metastasis of Tca8113 cells.
