Periodontal disease is the reason which caused periodontal tissue defects eventually leadto teeth missing.It’s the problem that how to prevent tissue from defects effectively and keepthe function.Tissue engineering brings the new technology.How to choose the ideal seed cellsand the meet the requirements scaffold material is the most importantObjective To establish a model of dog periodontal injured,therefore to evaluate thefeasibility of chitosan-silk fibroin-TCP as the scaffolds and the dog periodontal ligament cells(PDLCS)as seed cells of periodontal tissue engineering.Methods This study was carried out from May in 2010 to March in 2011. Anexperimental model was established in 6 dogs which were about 1-2 years old,the both firstmolar was used as the experimental tooth. They were randomly divided into 5 groups: controlgroup,chitosan,chitosan-silk fibroin-TCP. Each group was consisted of two. The dogperiodontal ligament cells cultured in vitro were collected. Then we take the FourthGeneration cells into the chitosan-silk fibroin-TCP. Then dogs were sacrificed after 8 weeks.The morphological changes were observed by HE staining.Cell proliferation was assayedusing MTT method.Results The result of HE staining shows that the Periodontal space reduced.Growth and proliferation of dog PDLCs of chitosan-silk fibroin-TCP group was notsignificantly different from that of the dog PDLCs of chitosan group.Conclusion Chitosan-silk fibroin-TCP showed excellent adhesion and goodbiocompatibility with the dog PDLCs in vitro compared with the chitosan group. It is feasible.
