The Expression And Significance Of Shh And Ptch Protein Of Hedgehog Signalling Pathway In Prostate Cancer

Prostate cancer (PCa)is the most common malignant tumor among men in WestonCountries, and it is the second cause of mortality in American males, just behind lung cancer.In China although the incidence of PC is less than Weston Countries, it is increasinggradually among population of weston life style and aging.In recent years, more and more evidence reveal that gene mutation may beresponsible for initiation of PCa. The role of Hedgehog (Hh) signaling parthway in theinitiation and progression of PCa has become research hot. Many researcher abroad considerthat Hh play a key role in the development of the prostate gland. Also,many studies reportedthat Hh play an important role in the PCa carcinogenesis and Metastasis. However themechanism of Hh parthway that promote the occurrence of PCa have not been fullyunderstood. It will have an important significance about that study on the structure andfunction changes of any factor of Hh in PCa. In this study we will investigate the expressionof Shh and Ptch protein in prostate cancer and analyse the relationship with pathologicalfeatures and explore its effect in generation and progression of prostate cancer.All data were from the pathology department of Prostate Diseases Prevention andTreatment Research Center of Jilin University.100cases of PCa biopsy specimens werestudied.Materials and methods:1. Serum PSA assaySerum PSA level was tested in all patients of the100cases of PCa, using ELISA assaykit (Sweden CanAg Company).2. BiopsySextant biopsies in prostate were performed on the patients using internationalstandards18G needle guiding by transrectal ultrasound. All specimens were routinely fixedin10%neutral buffered formalin and embedded in paraffin.3μm sections, HE staining.3. Histological observation and immunohistochemistry stainingImmunohistochemical staining of100cases was performed with the PV9000methodusing the monoclonal antibodies against Shh and Ptch.The results are as follows: (1) The expression of Shh and Ptch in prostate cancer were markedly elevatedcompared with their corresponding adjacent normal tissues，and the expression of Shh andPtch have positive correlation.(2) The expression of Shh and Ptch was related with Pca Gleason score（P <0.05）.(3)The expression of Shh and Ptch in prostate cancer have no relationship with serumPSA（P＞0.05）.The conclusion is as follows:Overexpression of Shh and Ptch of the Hedgehog signaling pathway in prostate cancer，and the expression of Shh and Ptch related with pathological grade，and have intimaterelevant with development and progression of prostate cancer，Shh、Ptch may becomemolecular targets of prostate cancer therapy.
