| Objective: We did a retrospective analysis on the clinical data of102cases who were diagnosed as MDS, comparing the chromosome abnormalpositive rate of bone marrow sample between conventional cytogenetics(CC)and fluorescence in situ hybridization(FISH), comparing the sensitivity ofthem.Methods:28cases of idiopathic thrombocytopenic purpura(ITP) or irondeficiency anemia (IDA) are randomly selected to set up positive thresholdfor FISH technology; the bone marrow cells of102MDS patients wereanalyzed with conventional cytogenetics(CC) and fluorescence in situhybridization(FISH). Besides the abnormalities of chromosome5,7,8and20,FISH were used to detect the loss of chromosome Y in65male patients and theabnormalities of12p in29cases.Results:(1) The total positive rate of FISH is obviously higher than CC(36.3%VS26.5%),statistical analysis of the positive rate of FISH and CCshows great significance(P<0.05),which indicates that FISH is more sensitivethan conventional cytogenetics(CC).(2) In the successful group of karyotypeanalysis, the positive rate of CC and FISH is40.3%and35.5%, respectively,there is significant difference between them (P<0.05). Except thechromosome7, the positive rate of chromosome5,8and20are improved byFISH.(3) In the failure group of karyotype analysis, the positive rate of FISH is37.1%, it is no significant difference comparing with the positive rate of FISHin the successful group of karyotype analysis (P>0.05).(4) The positive rate ofchromosome Y loss(-Y) is low, only1case with-Y is found by FISH in thefailure group of karyotype analysis.(5)there were2cases of29(6.9%)with abnormality of chromosome12by FISH.Conclusions: The sensitivity of fluorescence in situ hybridization (FISH)is significantly higher than the conventional cytogenetics (CC), and mini-clonalabnormality can be detected, FISH plays an important role in prognosis andcurative effect. But FISH is restricted by the type of probe, it can’t detect otherabnormal changes, so it cannot replace CC. From the paper,+8;20q-;-7/7q-and-5/5q-are the most common karyotype abnormalities in Jilin region. Thecombination of them can improve the positive rate of chromosome abnormalityin MDS patients. chromosome Y loss and abnormality of chromosome12areimportant in the course of MDS, the positive rate of them can be increased byFISH. |
