Objective:The aim of this study was to evaluate the inhibitory effects of Ginsenoside Rh2(G-Rh2)on proliferation, invasion and metastasis ability of oral squamous cell carcinoma (OSCC) cell line (KB). Its potential molecular mechanism was discussed.Methods:The survival and proliferate ratio of Kb cells treated by different concentrations of G-Rh2were measured by MTT assay exclusion test. The invade and migrate ability of Kb cells were measured by scratch wound assay and artificial basilar membrane rebuilteby matrigel respectively. To explore the anti-metastasis effect of G-Rh2, the activity of matrix metalloproteinase-2(MMP-2) of KB cells treated by G-Rh2was measured by gelatin zymography. The expression of matrix vascular endothelial growth factor (VEGF) of KB cells was measured by ELISA kit. DCFH-DA was used to measure intracellular reactive oxygen species.Result:(1) G-Rh2can inhibit the growth of Kb cells significantly in dose-dependent manner. When Kb cells were treated by10,20,40,60,80μg/mL G-Rh2, the inhibition rates were individually12.0%,6.4%,21.2%,30.2%,31.8%. When Kb cells were treated by10,20,40, μg/mL G-Rh2, invasion rates were65.4%,43.2%16.9%, and migrate rates were19.7%,10.5%and8.8%respectively. Therefore, migrate ability of Kb cells was obviously inhibited by G-Rh2.(2) G-Rh2can reduce the activity of matrix metalloproteinase-2(MMP-2), especially, MMP-2activity was completely inhibited when concentration of G-Rh2was40μg/ml. G-Rh2maybe eliminate the active oxygen within Kb cells, restrain the expression of VEGF, thus inhibit the formation of tumor blood essels.Conclusion:All the results demonstrate that G-Rh2plays an important role in the anti-metastasis and anti-angiogenesis of oral squamous cell carcinoma.
