Objective:To investigate the effections of new targeted therapy drugs Lapatinib on cell proliferation and cycle in human ovarian cancer cell line SKOV3in vitro and to explore its influence on the expression of HER2、P-HER2.Methods:Ovarian cancer cell line SKOV3were cultured in vitro under the irritation of Lapatinib with different concentrations (0、1、10、20、30and60μmol/L) for24h,48h and72h.The inhibition rates were detected by MTT array; the cell cycle of SKOV3cells exposed to Lapatinib was analyzed by flow cytometry; the expression of HER2and P-HER2was also examined by flow cytometry.Results:Lapatinib obviously inhibited the proliferation of SKOV3cells in a time and dose dependent manner, had the significant difference compared with the control groups (P<0.05); Different concentrations of Lapatinib made forG0/G1phase SKOV3cells significantly higher proportion (P<0.05), S phase and the G2/M issue of cell proportion significantly reduced (P<0.05);so before and after the application of lapatinib,the expression of HER2protein did not change significantly (P=0.13),and the expression of P-HER2protein had significantly different (P<0.05)Conclusions:Lapatinib can obviously inhibit SKOV3cell proliferation will be for the cell cycle block SKOV3in G0/G1phase;the mechanism has no obvious relation with the expression of the HER2,is closely related to downregulation of expression of P-HER2,and provides new targets and certain theoretical basis for its application in ovarian cancer. |
