BackgroundRheumatoid arthritis(RA) is a systemic autoimmune disorder that is characterized by chronic joint inflammation. Articular inflammationg causes activation of the synovial lining,expression of inflammatory cytokines,and T cell activation with autoantibody production.The main clinical manifestation includes symmetric, chronic and progressive arthritis which can destruct the articular cartilage and bone of the joints. The CIA model is similar to human RA in clinical,immunological and pathological changes.ObjectiveExamine the proliferative effect of synthetic cyclic human cartilage glycoprotein-39(HCgp39) on T cells of CIA rats,.determine the prevalence of interferon-γ(IFN-γ), interleukin-4(IL-4) and interleukin-17(IL-17) secreted by antigen specific T lymphocytes mentioned. Investigate the possible associations of the ant-HCgp39antibodies,the level of comp,joint inflammation,synovial phlogistic with clinical and laboratory indicators in CIA rats.Methods CIA was induced by SD rats with bCⅡ in completeFreund’s adjuvant. The T lymphocytes from CIA rats were isolated and incubated with HCgp39.The serum levels of IFN-y, IL-4and IL-17were measured with ELISA in CIA rats,the serum of comp, anti-HCgp39antibodies, joint inflammation,synovial phlogistic were examined at the same time.The correlation among them and HCgp39were compared.Results1. Comparison of normal rat all the CIA rats showed the inflammation and paw swelling,synovial membrane prolifeation and intense inflammatory cell infltration. The serum cartilage oligomeric protein (COMP) in CIA rats were elevated significantly compared with normal rat (P<0.01). In contrast to control group, HCgp39peptide group have manifested effect in splenocytes proliferation of CIA rats(P<0.01or P<0.05). And the HCgp39peptide group were significantly related with joint inflammation,synovial phlogistic and anti-HCgp39antibody,(P<0.01or P<0.05).2.The serum levels of IFN-y,IL-17and TGF-β were higher in CIA compared with the control, and there was statistical significance (P<0.01),but the levels of IL-4was lower than the control. The cell culture levels of IFN-y, IL-17, IL-4and TGF-β were higher in CIA compared with the control, and there was statistical significance (P<0.01)3.The percentage of HCgp39-AST cells in spleen lymphocyte in CIA was higher than that in the control,and there was statistical significance (P<0.01); In contrast to control group, anti-HCgp39antibody group and immune complex have manifested effect in splenocytes proliferation of CIA rats(P<0.01or P<0.05).ConclusionCIA was induced by SD rats with bCⅡ in completeFreund’s adjuvant. And the model was successfully established. The HCgp39peptide group have manifested effect in splenocytes proliferation of CIA rats,suggesting that maybe we can use HCgp39as a laboratory marker. The HCgp39peptide could induce Th cells secrete cytokines to participate in the pathogenesis of CIA.
