Study Of Chemical Constituents Of Caulis Mahoniae And Veratrum And Quality Control Of Caulis Mahoniae

This paper deals with the chemical constituents from Caulis Mahoniae and Veratrumdahuricum （Turcz.） Loes. F..This constituents were separated and purified by silica gel columnchromatography,Sephadex LH-20column chromatography and D101.Each compound was characterized by¹H-NMR,¹³C-NMR,HMBC,HMQC,EI-MS,ESI-MS et al.Fourteen compounds were separated,andeleven compounds were characterized.Caulis Mahoniae,the dried stem of Mahonia bealei （Fort） Carr.and Mahonia fortunei（Lindl.）Fedde,is belong to the f-amily berberidaceae.It has been widely used agains thygropyretic diarrhea,jaundice,conjunct-ival congestion and sore pain,gastropyretic toothache,carbuncle,dysentery and icterichepatiti-s.On the basis of chemical evidences and analyzingmodern chromatographic methods,three compunds have been isolated,and they are erthrosyringoylglyc-erol-4-O-β-D-glucoside（Ⅰ）,1-O-（β-D-glucopyranosyl）-（2S,3S,4R,8E）-2-[（2’R）-2’-hydroxytetracosenoilamino]-8-octadecene-1,3,4-triol（Ⅱ） and glucose（Ⅲ）.ⅠandⅡwere obtained forthe first time from Caulis Mahoniae.Veratrum dahuricum （Turcz.） Loes. F. is belong to Liliaceae family.Its roots areused as a Chinese folk medicine.It is coussin,poisonous and has effect of anemophlegmati-c vomiting,clearing away the heat of emetic,epilepsy seizures,jaundice,chronic malaria,heada-che,soer throat,mange,ulcer et al.On the basis of chemical evidences and analyzing moder-n chromatographic methods,eight compunds have been isolated and they are undecanoic a-cid,11-[[（2E）-3-（4-hydroxy-3-methoxypheny-l）-1-oxo-2propen-1-yl]oxyl]-2,3-dihydroxypropyl ester（L1）,3β-hydroxyisoandrosta-5,12（15）-dien-16-one（L2）,hexacosanoic acid2,3-dihydroxpropylester（L3）,Oxyresveratrol （L4）,verapatulin（L5）,syringaresinol（L6）,prosapogenin A of dioscin（L7）,daucosterol（L8）.L1and L2are new compounds,L3,L6and L7were obtained for the first time from the Veratrum.This paper established HPLC-UV fingerprinter method and content determinnationmethod for Caulis Mahoni,and evaluate the overall quality of different origin and batchesof Caulis Mahoni.Through the similarity calculation,clustering analysis and active content a -nalysis,realize contrastion of different source of Caulis Mahoni in Material base.The finger-print and content determinnation provide the scientific basis for drug safety and qualitystable controllable.Antitumor activity of12compounds from Caulis Mahoni and Veratrum dahuricum（Turcz.） Loes. F. were evaluated by the high content screening（HCS）.The result show that5compounds in the12affect function of transporter protein in the K562cell,and L5,L6,L11weresignificant（P<0.05）.The result provide experiment reference for the future to further study aboutimpacting function active of drug-resistant cell.
