Objective: To detect and analyze the expression and clinical significance ofplatelet-derived growth factor-AB (PDGF-AB) and thrombospondin-1(TSP-1), in healthypersons’ peripheral blood, clear cell renal cell carcinoma (ccRCC) tissue, cancer adjacenttissue, normal renal tissue and pre-operation peripheral blood of human clear cell renal cellcarcinoma patients.Methods: we collected35cases peripheral blood samples of healthy persons,47cases of ccRCC tissue,41cancer adjacent tissue,38normal renal tissue and35pre-operation peripheral blood samples of ccRCC patients. We used Enzyme-linkedimmunosorbent assays (ELISA) to determine the expression of PDGF-AB and TSP-1inthe three tissues extracts and peripheral serum samples, and analyzed their clinicalsignificances.Results:(1) The levels of PDGF-AB expression in ccRCC tissue, cancer adjacenttissue and normal renal tissue were32.79±19.91ng/ml,23.97±11.55ng/ml,14.73±7.77ng/ml respectively. They were successively decreasing. The expression ofPDGF-AB was higher in ccRCC tissue than cancer adjacent tissue and normal renal tissue(P=0.02, P<0.01). The expression of PDGF-AB was higher in cancer adjacent tissue thannormal renal tissue (P<0.01). The expression of PDGF-AB was positively correlated withthe TNM stage and Fuhrman grade of ccRCC (r=0.71,P<0.01; r=0.65,P<0.01).(2) Theexpression of TSP-1in ccRCC tissue, cancer adjacent tissue and normal renal tissue were36.44±4.80ng/ml,42.55±4.12ng/ml,46.55±1.55ng/ml respectively, which wassuccessively increasing. The TSP-1expression was lower in ccRCC tissue than canceradjacent tissue and normal renal tissue (P all <0.01). The expression of TSP-1was lower in cancer adjacent tissue than normal renal tissue (P<0.01). The expression of TSP-1wasnegatively correlated with the TNM stage and Fuhrman grade of RCC (r=-0.84, P<0.01;r=-0.70,P<0.01). The expression of PDGF-AB and TSP-1was no statistical significancewith the age, the sexual of patient’s and the size of tumor (P all>0.05).(3) There were nostatistical significance in PDGF-AB and TSP-1expression between patient’s peripheralblood sample and healthy person’s blood sample (2.96±2.39ng/ml vs2.75±2.01ng/ml,P=0.79;170.3±20.04ng/ml vs170.0±18.06ng/ml, P=0.62). The expression of PDGF-ABand TSP-1was no significant correlation in patient’s peripheral blood sample and ccRCCtissue (r=-0.14, P=0.43; r=-0.07, P=0.64).Conclusions:(1) The high expression of PDGF-AB may be important factor inpromoting tumor angiogenesis.(2) The low expression of TSP-1may be important factorin inhibiting tumor angiogenesis.(3) The expression level of PDGF-AB and TSP-1wasclosely related with TNM stage and Fuhrman grade in RCC which can be used as twoindicators to evaluate the development and grade malignancy of tumor.
