Objective: To investigate the expression of hypoxia inducible factor-1α (HIF-1α) and B7-H4inrenal cell carcinoma and para-tumor kidney tissue, and to study the hypoxic environment induced bycobalt chloride, renal carcinoma cell line786-O of HIF-1α and B7-H4expression changes in mRNAlevels and protein levels to explore whether the expression of B7-H4impacted in tumor tissue ofhypoxic microenvironment.Methods: Collected80cases of surgical resection of renal cell carcinoma specimens as theexperimental group, and the other75cases of para-tumor kidney tissue as the control group. Use themethod of cell crawling piece, Immunohistochemical and reverse transcription polymerase chainreaction to detect the protein and mRNA levels, the expression of HIF-1α and B7-H4, and collectedclinical data to analysis of the relationship between their expression levels and clinicopathologicalcharacteristics of renal carcinogenesis. In addition, renal carcinoma cell line786-O treated with Cobaltchloride to induce the hypoxic environment, and use the cells crawl piece immunohistochemistry andRT-PCR method to analyze the expression of HIF-1α and B7-H4.Result:1、HIF-1αexpression in renal cell carcinoma (45/80) is higher than normal renal tissues(2/75) next to the cancer, the difference was statistically significant (P<0.05); B7-H4in renal cellcarcinoma (42/80) is higher than in the cancer adjacent normal renal tissue (5/75), the difference wasstatistically significant (P<0.05). There was no statistically significant correlation between HIF-1α andB7-H4expression and gender, age and tumor size (P>0.05). There was no statistically significantcorrelation between B7-H4and HIF-1α in lymph node metastasis (P<0.01).2、compared with Renal cellcarcinoma and adjacent normal renal tissue, the expression of B7-H4and HIF-1α mRNA expressionincreased (P<0.05).3、After renal carcinoma cell line786-O treated with Cobalt chloride to induce thehypoxic environment, HIF-1α and B7-H4mRNA and protein levels were up-regulated.Conclusion: HIF-1α and B7-H4have different levels of highly expressed in renal cellcarcinoma, and both the expression of closely related, and cobalt chloride can increase HIF-1αandB7-H4expression in the786-O, indicating that tumor hypoxic microenvironment can induce theexpression of B7-H4. |
