ObjectiveLight-microscopy different month-stages of the trend of histophological changes in SD rats ovarian (follicle, The corpus luteum counting of estrogen cycle, interstitial corpus luteum glandsã€Germinal epithelial) and uterus (the uterus counting of Estrogen cycle), statistical analysis of different month-stages of the law of histomorphological changes on ovarian and uterus, provide basic data for drug safety evaluation;Prevent the misdiagnosis (mistaken the natural aging for causing by drug toxicity lesions) happening in long-term toxicity tests; Understand the domestic existing conditions in raising SD rats female reproductive system characteristics of natural aging, and compared with the foreign existing material of raising SD rats female reproductive system characteristics of natural aging; For traditional Chinese medicine/western medicine anti-aging research parameter selection to provide reliable and quantitative/half quantitative basis.MethodsSD rats, female, SPF, a total of40pes, full nutrition feed breeding, free food, and not to any treatment. After3monthsã€14monthsã€17monthsã€18months, take10rats only were anesthetized with chloral hydratebe, then killed with fracturing their cervical vertebra.The uterus and ovary were removed en bloc. Extraneous tissue was trimmed away under a dissecting microscope. Uterus and ovaries were then fixed in10%neutral buffered formalin. After24hr, all tissues were removed from formalin andstored in70%ethanol, followed by embedding in paraffin,sectioning at5microns, and staining with hematoxylin and eosin. For histopathologic assessments, transverse sections were taken through the vagina and one uterine horn of females.All sections were examined by a board-certified veterinary pathologist for any histologic abnormality. Greatest cross-sectional diameter of the uterus and ovaries were measured using a calibrated stage micrometer. The average of atrefic follicle in ovary tissues gradually increases with months increasing, but the overall difference among the groups were not statistically significant (P>0.05). The average of corpus luteum totality in ovary tissues gradually decreases with months increasing, and the overall difference among the groups were statistically significant (P<0.001).
