Objective:The objective of this thesis is to provide preliminary experimental basis for pharmacological mechanism and clinical application of compound huo shui tab and root and leaf extracts of Rosa bracteata Wendl in treatment of iron overload.Methods:First, iron overload rat models were made. A part of the models were intraperitoneally injected Iron Dextran injection in high, medium or low dose, respectively, while the rest were intraperitoneally injected Iron Dextran injection in low dose and fed Polysaccharide-Iron Complex Capsules in the meantime. According to differences in the models’growth tendency, organ coefficient and serum iron content, the best molding method was screened out. Afterwards, models made in the best molding method were fed compound huo shui tab and root and leaf extracts of Rosa bracteata Wendl. Their general condition, growth tendency, and some indicators related to iron metabolism, including contents of serum iron, ferritin, transferrin and transferrin receptor, were observed and recorded. At last, all data were statistically analyzed by SPASS17.0.Results:1. The best molding method was screened out. The serum iron content of rats which were kept feeding SPF full feed nutrition and every second day intraperitoneally injecting150mg/ml Iron Dextran injection for four weeks became about three times that of normal rats.2. Significant differences in contents of serum iron, ferritin, transferrin and transferrin receptor were observed between rats fed leaf extracts of Rosa bracteata Wendl and the model control group (P<0.05). In contrast, no statistic significance was found in the conparison between rats fed compound huo shui tab and the model control group (P>0.05). However, rats fed compound huo shui tab had obvious increases in appetite and weight, which probably was the effects of the large amount of flavonoids in the compound. In addition, no statistic significance was found in the conparison between rats fed root extracts of Rosa bracteata Wendl and the model control group (P>0.05)Conclusion:1. The best molding method is keeping feeding SPF full feed nutrition and every second day intraperitoneally injecting150mg/ml Iron Dextran injection for four weeks.2. The research illustatrates that leaf extracts of Rosa bracteata Wendl have positive effects on reduction of iron overload rats’serum iron content and protection of organism, which is related to the large amounts of chelate iron and Fisetin contained. Compound huo shui tab is helpful in improving general symptoms, increasing weight, and providing protection to some extent, but has little influence on iron metabolism. Whether this is related to the antioxidation activity of flavonoids contained in the compound is to be determined in further research.
