Enterovirus71(EV71) belongs to Enterovirus, Picornaviridae, with asingle-stranded small RNA. It is very infectious, pathogenic and harmful particularlyto the central nervous system. In recent years, EV71is very attractive to publicbecause of its risk. It was found that this virus is one of the major pathogens ofHFMD(children’s hand, foot and mouth disease, HFMD), can lead severe HFMDespecially to children under6years old. According to statistics, more than90%ofsevere HFMD cases were caused by EV71. In addition to the typical symptoms ofHFMD, there were up to11.1%and0.6%of children patients suffering from centralnervous system diseases, for example aseptic meningitis and encephalitis. Patientcondition deteriorated rapidly. Mortality rates were higher than250millionths inrecent years. An outbreak of EV71is serious harmful to the health of children, andpsychological panic to public. The vaccine or cure against EV71has not yetdeveloped in currently. Prevention of HFMD mainly rely on the establishment ofgood health habits. So, the development of safe, efficient and economical vaccineto EV71is imminent.The objective of this study is the development of EV71epitope peptide vaccine.The gene sequences of29C4strains(from Fujian, Fuyang, Anhui, Jiangsu,Zhejiang and other provinces), and28the NCBI gene bank B type EV71strainsfrom Genbank were alignmented.5high homologous epitope were found in thedifferent serotypes of the virus. With DIII protein of Japanese encephalitis as carrierprotein, the five epitopes was reorganized into eight kinds in the way of T-T-B andT-B-B series,8epitope peptide vaccines were designed on the basis of prokaryoticexpression system.Immunogenicity trials of the recombinant protein showed that the antibody levelsin the sera of mice increased significantly. Type C, E, F, H can induce higher levels of antibody. These results lay the foundation for new vaccine design anddevelop the value of EV71epitope peptide vaccine.
