A significant increase of apoptosis cells was observed at MYCNsiRNA group compared with those at control siRNA group.Conclusion:The bone marrow and bone xenograft mouse model can be successfullyestablished by direct local injection of LA-N-5cells and it would be a valuable tool forunderstanding the biology of metastatic neuroblastoma to bone and marrow, andevaluating the efficacy of potential therapeutic agents. MYCN siRNA was highlyconcentrated in neuroblastoma tissue via folate-nanoliposome entrapment afterintravenous injection. It significantly reduced MYCN mRNA expression and inhibitedneuroblastoma cell apoptosis in nude mice, these findings suggest that FR-targetedliposome is an effective vehicle for tumor-targeted siRNA delivery and the liposomeentrapped folate FT-targeted MYCN siRNA could be a novel therapeutic approach forneuroblastoma.
