Objective:To create a post-traumatic stress disorder in an animal model on the basis of the different stress intensity and explore the reasons for stress-sensitive, to explore whether this stress sensitization is due to changes in the ability to learn after the traumatic event.METHODS:The experiment1,selected72male SD rats, using formalin injection and foot shock stress mode and single prolong stress (SPS) to establish post-traumatic stress disorder animal models, the use of startle response box, elevated plus maze experiments, social aversion test procedures for post-traumatic stress disorder symptoms.Experiment2,selected48male SD rats, animal model of formalin injection and foot shock to the establishment of post-traumatic stress disorder, scared startle response box and inhibitory avoidance test, test period of acute stress in rats with non-Lenovo learning (neutral, negative stimuli). Using the elevated plus maze test, social aversion test program behavior test for post-traumatic stress disorder symptoms.Experiment3, selected48male5D rats, using the formalin injection and the foot shock was built animal models of post-traumatic stress disorder, the use of food location preference for program testing in rats with acute stress of non-associative learning (positive). Formalin foot shock group and control group in the hyperarousal symptoms significant difference (t=53.82, p=0.001). On the avoidance symptoms of behavioral tests, the formalin group in the open arm time (p=0.009) and frequency (t=0.185, p=0.008) was significantly less than the control group. Social avoidance symptoms,Formalin foot shock group into the social time region (t=23.00, p=0.019) and frequency (t=1.962, p=0.009) than the control group.Experiment2, early in acute stress period stimulation has nothing to do with the trauma, stress group than the control group, but between them there is no significant difference (t=0.477, p=0.638). Late in acute stress period, between the control group and stress group, there is a significant difference (t=3.126, p=0.005). Early in acute stress period negative learning ability tests, the stress group and control group compared to the incubation period is longer, but the difference was not significant (t=-1.264, p=0.230). Late in acute stress, between control group and stress group, there were significant differences (t=-2.337, p=0.032). Late in acute stress period the lower stimulate(90dB) is high correlated with avoidance symptoms (r=0.455, p=0.026). Learning ability of acute stress of the late negative test values and social avoidance test data are highly correlated (r=-0.553, p=0.029).Experiment3, acute stress period,there was no significant difference between the control group and stress group in the positive learning ability. Positive learning ability had and no significant correlation with avoidance symptoms, social avoidance symptomsConclusion:1) Formalin foot shock model of PTSD animal model with high face validity.2) Rats of PTSD in the acute stress of stimulation has nothing to do with the trauma over time change, and enhance.3) PTSD in rats to study the sensitivity to negative stimuli in the acute stress of growing over time.4)Rats of PTSD between the sensitivity of acute stress on the learning of positive stimuli and normal rats no significant difference.5)Late acute stress perior of low intensity and trauma unrelated to stimulus-response can predict the avoidance symptoms of PTSD.6)Late in acute stress perior the learning sensitivity to negative stimuli could predict PTSD symptoms of social avoidance.
