Objective: To study the relationship between the exon149A/G polymorphism of thecytotoxic T lymphocyte associated antigen-4(CTLA-4) gene and the response toantithyroid drugs(ATD)in Graves’disease (GD).Method: The A/G polymorphism of CTLA-4gene was tested by PCR-RFLP in102patients with relaplse Graves’ disease,including60cases with early relapses within3years and42cases with relapses were more than3years after withdrawal ofantithyroid drug(ATD),40initial GD patients and80healthy controls were includedin the study.Results:1. The frequencies of GG genotype and G allele were32.5%and58.8%ininitial GD patients,40.0%and67.2%in patients with relaplse GD,12.5%and35.0%inhealthy controls. There was distinct difference of GG genotype and G allelefrequencies between all GD patients and healthy controls(P<0.05).2.The frequencies of GG genotype and G allele in patients with relaplseGD(40.0%and67.2%) were more than the initial GD patients (32.5%and58.8%).However,the differences were no statistically significant(P>0.05).3. Further analysis, the frequencies of GG genotype and G allele were53.3%and74.2%in early relapses group(group A),21.4%and57.1%in the3yearsafter withdrawal of antithyroid drug(ATD) relapses group(group B). There wasdistinct difference of GG genotype and G allele frequencies between group A and theinitial GD patients(P<0.05),but no significant difference between group B and initialGD patients(P>0.05).Conclusion:1. CTLA4gene exon149A/G polymorphism is associated withGraves’disease. 2.The GG geneotype of the exon149of CTLA-4gene may be a riskfactor of the bad curative effect of ATD or early relapsed....
