Basic Study Of Pemetrexed Chrono-chemotherapy Tumor

Objective:1. To construct the model of the nude mice bearing lung adenocarcinomacell line A549cells, which laid the groundwok for studying tumor-suppressing rates.Pemetrexed disodium was given to the node mice at different times in the day and nightaccording to the theory of chronopharmacology.2. The adverse reactions ofchemotherapy according circadian rhythm was studied on the model of Balb/c nudemice.3. The expression of TS mRNA and TP mRNA in transplant tumor cells wasdetected by qRT-PCR accordingly.Methods:1.1x107A549cells in exponential phase of growth were vaccinated in thegood health body of Balb/c nude mice by subcutaneous injection(injection under theskin).2. The tumor tissue was measured from major axis to minimum width every otherday, and calculated the volume for drawing the growth curve.3. As the tumor tissuegrowing to meet the volume of0.5-1.5cm3, intraperritoneal injection with pemetrexeddisodium was performed immediately. Meanwhile, The observiation was carried out asthe following: the appetite, the movement, and the mental state of nude mouse, as wellas the tumor size. The weight of the nude mice was measured twice a week for drawingthe chart. Complete blood count was detected by wiping out of the mouse tail at thetime of the day8and12.4.by the end of the experiment, the tumor weight of the nudemice was calculated for counting the tumor inhibition rate.5. qRT-PCR was applied todetect the expression of the TS mRNA and TP mRNA in the tumor tissue.Results：Pemetrexed for lung adenocarcinoma nude mice transplant tumor has goodcurative effect（P<0.05），7h group tumour inhibition rate was the highest（P<0.05），the slowest Tumor growth（P<0.05），the more nude mice weight（P<0.05），15h grouptumour inhibition rate was the least（P<0.05），the fastest Tumor growth（P<0.05），the least nude mice weigh（tP<0.05），relative express level of TS mRNA of7h group tumortissue was the lowest，however，TPmRNA is the highest. Difference was statisticallysignificant（P<0.05）.Conclusion：Pemetrexed for lung adenocarcinoma nude mice transplant tumor haveapparent inhibition and efficacy and side effects appear hour rhythm changes，thecurative effect of7h group was the best. Preliminarily thinking， it look on TS mRNAand TPmRNA as side effects and curative effect Biological marker of pemetrexed， itprovided us reference For rationalization and individualized clinical application ofpemetrexed.
