The green fluorescent protein(GFP) is a kind of luminescence protein, which can catalyze itself into chromophores and emit green fluorescence when exposed to blue light or ultraviolet light. Green Fluorescent Protein whose molecular mass is stabl,convenient and non-harmful to the cells, so it has been used widely as a gene label in the field of biology. The fluorescence of EGFP-transgenic mice can be expressed in whole-body and it can be highly observed in major organs including brain, heart, lungs, liver, kidney, intestine, stomach and pancreas.By crossing the BALB/c male nude mice with the EGFP-transgenic C57BL/6J female mice, the EGFP-transgenic nude mouse is obtained.The EGFP-transgenic nude mouse is a very useful tool mouse which can be used to track the taget cell by the technology of optic imaging. It can also be used to research the sterm cell and cancer cell. The following points are what we do in our reaserch:1. The EGFP-transgenic nude mouse implanted A549cellsThe EGFP-transgenic nude mouse was obtained by crossing BALB/c male nude mice with the EGFP-transgenic C57BL/6J female mice. Using methods of gross anatomy,we can inspect the growth of thymus. The fluorescence imaging of whole-body and organs can verify the expression of green fluorescent protein.Six-week-old EGFP-transgenic nude mice were subcutaneously injected with red fluorescent protein-expressing human A549lung carcinoma cells(RFP-A549) with a single dose of2×106, testing and verifying the immunodeficiency of EGFP-transgenic nude mouse through the growth of tumor. A model of EGFP-transgenic nude mouse implanted RFP tumor was established to evaluate the whole effect of Dual-color fluorescence imaging, then the source of cell and vessel was distinguished, the tumor metastases were detected through Dual-color fluorescence imaging.2.The location of human umbilical cord mesenchymal stem cells(hUC-MSCs)The uMSCs are separately labeled with RFP and luciferase(LUC). The EGFP-transgenic nude mice were implanted with human umbilical mesenchymal stem cells (RFP-hUC-MSCs and LUC-hUC-MSCs) through the tail vein, researching the distribution and extinction of the human umbilical mesenchymal stem in vivo imaging. We collected data at differernt time from same experimental subject through to observe the survival and migration of uMSCs in vivo optical imaging, so that we can eliminate individual difference and get more credible data.The liver of mice was injured by CCl4, the injured liver was observed by immunohistochemistry under the condition of accepting uMSCs. Meanwhile,we compared the imaging effect of RFP with that of LUC.
