Objective:To detect the expression of Slit2/Robo singal pathway onrat’s VSMC, and to observe the effects of migration of VSMC and theexpress of its receptor Robo after stimulated by exogenous Slit2protein.Method: VSMCs were primary cultured and identified.ModifiedBoyden chamber way was used to detect migration of VSMC.In this way,Slit2and PDGF-BB with different concentions were added into the lowerchambers. CKK-8way was used to detect proliferation of VSMC.RT-PCRwas use to detect the gene`s expression of Slit2and robos in VSMC and thedifference of gene robo stimulated by Slit2.Immunofluorescence was used todetect expression of Slit2/robo proteins in VSMC.Result: All genes and proteins called Slit2,robo1,robo2,robo4areexpressed in rat’s VSMC,however the gene of Robo4expresses weakly.Although exogenous Slit2have no effect on the proliferation of VSMC, itinhibites VAMC`s migration even induced by PDGF. After stimulated byexogenous Slit2protein, genes express of robo1/bobo4are upregulated,andgenes express of robo2are downregulated. Conclusion: Migration of VAMC can be prevented by exogenous Slit2protein,and expression of Robo can be influenced by prevented byexogenous Slit2protein too. Through the research on Slit2/robo singalpathway,it probably can provide a new treatment target on antherosclerosisand the therapy for restenosis after PTCA/PCI.
