Objective:To investigate the effects of PI3K-AKT signal pathway on the proliferation, apoptosis and invasion of colon carcinoma cells (SW480),and to explore its possibility mechanism.Methods:SW480cells lines were cultured with different concentrations of LY294002and observe the results, researching the effects of the vitality of cells caused by PI3K-AKT signal pathway by means of MTT as say; detecting the apoptotic rate by Flow Cyto metry (FCM); Transwell invasion test was used to examine the invasion ability of the SW480cells; The protein level of VEGF and MMP-9were detected by Western bloting.Results:LY294002inhibits the growth of SW480cell lines, induce apoptosis; inhibition the PI3K-AKT signal pathway, the invasion of SW480cells degrades and the expression of VEGF and MMP-9reduces.Conclusion:PI3K-AKT signal pathway plays an important role in the proliferation, apoptosis and inhibition of colon carcinoma cells (SW480). Its mechanism may be related to inhibiting the VEGF and MMP-9. PI3K-AKT signal pathway probably is the potential target of therapy in treating the colon carcinoma.
