ObjectiveSpermatogenesis is the main process in the male reproductive.Proliferate of the Spermatogonia ensure the progress of spermatogenesis.With the growing age, Sperm quantity and quality are declining. mTORsignal transduction pathway plays a central regulatory role in cellproliferation and differentiation.So, mTOR signaling pathway plays animportant role in the spermatogenesis. Detect the expression of mTOR in therat testis, and to explore its relationship with sperm. Lay an importantfoundation for further study of specific mechanisms.MethodsHistological location of mTOR by immunohistochemical, study theexpression levels of mTOR in the sexually mature male rats in SD heart,liver, kidney, brain, testis tissue by RT-PCR.Fluorescence quantitative PCR, western blot to detect the expression ofmTOR in the testes of male SD rats of different developmental stages.Toobserve the morphology changes of the SD rat testis in different developmental stages by HE staining.To build rapamycin SD rat model,and detect the expression of mTORmRNA in the normal group, rapamycin group and control group byFQ-PCR.To detect the expression of mTOR protein and mTOR downstreamtarget proteins P-P70S6K/P70S6K by using Western Blot.Results:1、The immunohistochemistry results showed that the mTOR-positiveimmune staining was mainly located in the cytoplasm of the spermatocytesof the testicular seminiferous tubules and a small amount of primaryspermatocytes.2、Compared with other major organs of the mature SD rats mTORexpression in the testis is relatively high. With the increase in SD rats age,the expression of mTOR in the testis decreased, and accompanied byvacuolization of the seminiferous tubules;and mTOR downstream target theprotein P70S6K phosphorylation efficiency decreased.3、treat SD rat with rapamycin,and observed that vacuolization of theseminiferous tubule in the testis is similar to the old rats,and reducedefficiency of phosphorylation of mTOR downstream target proteinsP70S6K.Conclusion:1、 The first time, we found that mTOR expression in the ratseminiferous tubules spermatocytes. 2、With the growing age, because of the mTOR expression and itsphosphorylation of downstream target proteins P70S6K decreased.the countof rat sperm reduced and seminiferous tubule vacuolization increased3、mTOR contributing to spermatogenesis through phosphorylation ofP70S6K and P-P70S6K promote spermatogonial cell proliferation andprotein synthesis.
