Objective To explore the expression of ATP-binding cassettetransporter G2(ABCG2) and hypoxia-inducible factor-1alpha(HIF-lα) inhuman hepatoma SMMC-7721cells under the anoxic conditions and theirrelationship.Methods SMMC-7721cells were divided into control group,24hhypoxia group,48h hypoxia group and72h hypoxia group. Western blotwas employed to detect the protein expression of ABCG2and HIF-lα inSMMC-7721cells of each group. SMMC-7721cells in72h hypoxia groupwere treated with HIF-1α inhibitor YC-1at the finial concentration of1.0,2.0and4.0μmol/L, respectively, and Western blot was used to inspect theprotein expression of ABCG2and HIF-lα in SMMC-7721cells treated bydifferent concentration of YC-1.Results With prolonged hypoxia, ABCG2and HIF-1α proteinexpression were gradually increased(P<0.05), and the positive correlationwas found between the expression of ABCG2and HIF-1α(r=0.944,P<0.05).The protein expression of ABCG2and HIF-1α weredown-regulated with increasing concentration of YC-1(P<0.05).Conclusion ABCG2and HIF-1α expression are increased with increasing hypoxia, and ABCG2up-regulation may be one of themechanism of HIF-1α-mediated survival and drug resistance ofHepatocarcinoma cells under the anoxic conditions.
