ObjectiveTo analyze the80SNP loci of14genes about BRAF、RET、MEN1、FOXE1,ect and to study the value of these genes in the thyroid carcinoma.Methods1.80SNPs loci were selected according to the information provided by90PUBMEDS from NCBI、OMIM and KEGG, standard:1)related genesof development mechanism and its recurrence/transfer or lossdifferentiation in PTC/FTC;2) related genes of diagnosis, treatment andprognosis inTC.2.According to the criteria, we selected80SNPs of14genes from77PUBMEDS, including:36SNPs of BRAF and RET in PTC;7SNPs ofRAS and PAX8-PPARγ in FTC;26SNPs of TP53and CTNNB1in pDTC;and11SNPs of other related genes.3.We collected11cases of TC,including8cases of PTCs,2cases of FTCsand1case of pDTC and22cases of TA.The tissues include carcinoma andbeside carcinoma.We must put these tissues into the liquid nitrogen to frozen in30minutes,and then move these tissues into the fridge of80degrees below zero after48hours.4.These SNPs were typed by Sequenom MassARRAY.To valuation theclinic application of80SNP loci using the fresh sample of thyroidcarcinoma.Results1.we found the expression of80SNPs in TC and TA,except rs1733832of BRAF.2.There were the significance of difference in rs669976of MEN1andrs965513of FOXE1between tTC and TA.3.The mutation of rs722494from TP53detected the significance ofdifference between the TC and TA.4.The incidence rate of the fusion type of80SNPs were47.5%、7.5%and8.75%in PTC、FTC and pDTC; The incidence rate of the deletion typeof80SNPs were2.5%and1.25%in PTC and FTC.ConclusionWe study the important role of80SNPs loci in the thyroid neoplasms bythe method of Sequenom MassARRAY.The introduction of this study intoclinical practice could provide useful information of11genes for thediagnosis and possibly for the prognosis and treatment of TC in thefuture.
