ObjectiveTo investigate whether7-difluoromethyl-5,4’-di-n-octylgenistein (DFOG)suppresses the growth and induces apoptosis in human lung cancer A549cell line andits mechanisms.MethodsHuman lung cancer A549cells were cultured in vitro. The cell viability wasevaluated by MTT assay. The ability of cell colony formation was detected by thecolony formation on plate. The cell cycle phase distribution and percentage of sub-G1cells (apoptotic rate) were determined by flow cytometry (FCM) with propidiumiodide (PI) fluorescence staining. The level of histone/DNA fragment in A549cellswas measured using enzyme linked immunosorbent assay (ELISA). The level ofMcl-1protein expression was analyzed by Western blot.ResultsMTT assay indicated that DFOG inhibited the viability of human lung cancer A549cells, in a concentration-dependent manner, with IC50value was3.9±1.2μmol/L. Theclonogenic Assay indicated that DFOG (2.0,4.0,8.0μmol/L) significantly inhibitedthe anchorage-dependent growth. FCM analysis with PI stainning indicated thatDFOG resulted in cell cycle arrest in G2phase and efficiently induced apoptosis ofhuman lung cancer A549cells. ELISA indicated that DFOG increased the level ofhistone/DNA fragment in lung cancer A549cells, in a dose-dependent manner. Western blot analysis indicated that2.0μmol/L DFOG reduced the level of Mcl-1protein expression, in a time-dependent manner.Conclusion1. DFOG possesses the effects of inhibition of viability and anchorage-dependentgrowth in human lung cancer A549cell line, in a concentration-dependent manner.2. DFOG effectively induces apoptosis of human lung cancer A549cells, in aconcentration-dependent manner.3. The induction of apoptosis by DFOG in human lung cancer A549cells is involvedin down-regulation of the expression level of Mcl-1protein.
