Effects Of β3-adrenoceptor Agonist BRL37344on Differentiation In Preadipocyte

Objective:To investigate the effect of β3ARa BRL37344on the differentiation in preadipocytes, and to explore the possible mechanism.Methods:3T3-L1preadipocytes and Human primary preadipocytes were cultured respectively. Cells were induced differentiation after they were adherent and reached confluence2days. Preadipocytes were cultured in vitro and differentiated by four groups of inducers:control, basic inducers, BRL37344, basic inducers+BRL37344. Cells were stained by oil red O, and mRNA levels of PPARy2、aP2、LPL in adipocytes during differentiation were detected after the intervention of three different doses (10-9,10-8,10-7mol/L) of BRL37344. Then cells were intervenced by10-7mol/L of BRL37344. Cells were also stained by oil red O, and mRNA levels of PPARγ2、aP2、LPLin3days(0、2、8) during adipocytes differentiation were detected.Results:(1) mRNA levels of PPARγ2、aP2/LPL were dose-dependently down-regulated by BRL37344in3T3-L1preadipocytes. The maximum inhibition of PPARγ、aP2、LPL mRNA levels were observed after treatment with10-7mol/L BRL37344for8days (P<0.05).(2) The mRNA levels of adipocyte differentiation related genes above-mentioned were increased linearly in three different days (0、2、8d). But they were down-regulated compared with the control group after exposure to10-7mol/L BRL37344(P<0.05). So were the mRNA levels of the genes above-mentioned after exposure to induced differentiation+10-7mol/L BRL37344compared with induced differentiation group (P<0.05).(3) Primary culture of human preadipocyte was a failure maybe because of the mixture of the DMEM/F12medium.Conclusion:(1) We successfully established the differentiation model of3T3-L1 preadipocytes.(2) β3-adrenoceptor agonist BRL37344could inhibit adipocyte differentiation in3T3-L1preadipocytes.(3) BRL37344could down-regulate the mRNA levels of PPARγ2、aP2、LPL in adipocyte differentiation in3T3-L1preadipocytes, which maybe the mechanism of it.
