Toxoplasma gondii is an apicomplexan parasite that infects all warm-blooded animals,including humans. Infections of Toxoplasma gondii may cause encephalitis or chorioretinitis in immune-compromised hosts, and can result in miscarriage or neonatal malformations with congential infection. There is no effective vaccine to resist T.gondii infections up to now, so a new kind of cheap and effective vaccine is needed urgently. Rhoptry protein18(ROP18)can cause the proteins of hosts lose efficacy, thus protect the membrane of T.gondii. Apical membrane antigen-1(AMA1) is a microneme protein and it is very important in the process of T.gondii invasion. In this research, we amplify the specific genes AMA1and ROP18using PCR method, and then construct eukaryotic recombinant plasmid pBudCE4.1-AMA1-ROP18and express in human foreskin fibroblast (HFF) cells. Then identify the plasmid with SDS-PAGE and beta-actin. After vaccing the BALB/c mice, we found these vaccines could induce immunological response of mice, and could protect mice from T.gondii RH infection in a certain extent. these results establish a foundation for the future research on DNA vaccine.
