Effects Of Quercetin On The Biological Behavior And VEGF Protein Expression On Human Renal Cell Cancer Cell Line ACHN

Objective: To study the effect of quercetin on the proliferation、migration and invasionon human renal cell cancer cell line ACHN， to study its probable molecularmechanisms.Methods：ACHN cells cultured in vitro，with different concentrations of quercetintreated cells treatment，the inhibition of proliferation of ACHN cell was studied bymethyl thiazolyl tetrazolium (MTT) assay, the abilities of cell migration were detectedby wound healing assay, the abilities of cell invasion was evaluated using Transwellchambers，the expression of vascular endothelial growth factor (VEGF) were detectedby Western blot assay.Results：After quercetin treatment, the proliferation of ACHN cells were obviouslydecreased, which showed a dose-and time-dependent manner(P<0.05); the abilities ofmigration of quercetin treated group，compared with the blank control group，wasdecreased; the abilities of invasion of quercetin treated group were suppressed,compared with the blank control group. Quercetin down-regulation the expression ofVEGF protein in a dose-dependent manner. Conclusion：Quercetin can inhibit the abilities of cell proliferation、migration andinvasion of ACHN cells； with different concentrations of quercetin treateddown-regulating VEGF expression on human renal cell cancer cell line ACHN. Theeffect of quercetin on the proliferation、migration and invasion in renal cancer cell，themechanisms may have close relationship with down-regulation of expression of VEGF.
