Objective:①To successfully copy the model of explosive brain injury in rabbit,②To investigatethe changes of caspase-3、cytochrome C、bax and bcl-2in brain tissue with explosive braininjury in rabbit, and the Influence of early hyperbaric oxygen treatment on theexpression of them.③To investigating the effect of early hyperbaric oxygen treatmentwith explosive brain injury in rabbit and possibly related mechanism, providing atheoretical basis with the application of early hyperbaric oxygen treatment on theclinical.Method:①150New Zealand white rabbits, weighing2.0~2.5kg, were randomly divided intocontrol group(n=10),model group(n=70) and hyperbaric oxygen treatmentgroup(n=70).After successful modeling,the model group and the hyperbaric oxygentreatment group was subdivided into7groups,namely groups1h,6h,12h,24h,3d,7d,14d after injury, according to sacrifice time point and each group had10rabbits.Explosive brain injury model was established by paper detonator in the model groupand the hyperbaric oxygen treatment group,no explosive injury was performed incontrol group, than using early hyperbaric oxygen therapy to intervene the hyperbaricoxygen treatment group. The expression of caspase-3、cytochrome C、bax and bcl-2ineach rabbit brain were observed immunohistochemical after making the specimen with brain, and use statistical software to statistical analysis.Results:①The expression of caspase-3in the model group increased at1hours after injury andreached peak at24hours, and then decreased(p﹤0.05), the expression of caspase-3was also obvious at7days; Besides14days, the changes were significantly diferentbetween control and model groups(P<0.05); Compared with model group,besides14days, the expression of the caspase-3in hyperbaric oxygen treatment group decreased(P<0.05).②The expression of cytochrome C in the model group increased at1hoursafter injury and reached peak at12hours,and then decreased at24hours(P﹤0.01).Compared with model group,cytochrome C expression of the hyperbaric oxygentreatment group decreased at Early at1h,6h,12h,24h after injury (P﹤0.05);③Compared with control group,the expression of Bax and Bc1-2in the model groupincreased at1h,6h,12h,24h after injury(P﹤0.05). Compared with model group,theexpression of Bax in the hyperbaric oxygen treatment group decreased at1h,6h,12h,24h after injury(P﹤0.05), the expression of Bc1-2in the hyperbaric oxygentherapy group increased at1h,6h,12h,24h after injury(P﹤0.05).Conclusions:The expression of cytochrome C in nerve cells of the brain increased may be key stepswith protecting the nerve cells in the cerebral cortex from apoptosis at1h,6h,12h,24hafter brain explosive injury in rabbits. At1h,6h,12h,24h after early explosionhyperbaric oxygen treatment, the expression of caspase-3、cyt C and bax all decreased,and the expression of bcl-2increased, therefore,the effect of early hyperbaric oxygentreatment can induce the expression of Bc1-2protein and decrease the release of theexpression of Bax, thereby prevent cytochrome C from mitoehondria to protect brain.
