| With silk fibroin as drug carrier, a w/o/w doubLe emulsion methord was developed to prepare silk fibroin sustained-release microspheres through connecting aspirin.Besides,cellulase hydrolysis extraction and purification of safflor yellow was studied.The study contains: preparation of regenerated silk fibroin, preparation of aspirin silk fibroin sustained-release microspheres and their quality evaluation,studies on celluase hydrolysis extraction and purification of safflor yellow and preparation of safflower microspheres. Effects of emulsifier, the concentration of silk fibroin solution, the ratio of oil and water phase as well as the proportion of aspirin and silk fibroin solution were researched on the preparation and properties of silk fibroin microspheres. Silk fibroin drug-loading microspheres and encapsulation efficiency were determinated by ULtraviolet spectrophotometric.Drug release properties of microspheres was evaluated by release in vitro. With the content of hydroxysafflor yellow A as index, cellulase concentration, extraction time and temperature, as well as pH were studied on the effect of safflower yellow extraction. The optimization of cellulase was selected by means of orthogonal test. Through orthogonal test and single factor test,the concentration of ethanol, stirring speed,and pH of the solution affect of the purity and transfer rate of hydroxysafflor yellow A by use of alcohol precipitation process. After purification, the purity of HSYA can reach above50%. |
