Objective: Our aim is to detect the relationship between5-HT receptors geneexpression changes in hippocampus of model rat with liver-qi depression, which givesa preliminary discussion on the pathogenesis of depression emotion, as well as studiesthe effects of Shuyu capsule&Radix Stellaviae.Methods: After being chosen,liver-qi depression model rats were prepared by usingchronic mild stress (CMS)method. And then the rats were evaluated by sucrosepreference test and open field test. We used fluoxetine as masculine medicine,detected the5-HT receptor gene expression changes in hippocampus of rats in thegroups including the normal group, liver-qi depression model group, and the threetreatment groups by using gene chips and real-time quantitative PCR.Results: Compared with the normal group, the total score of open field test of rats inliver-qi depression model group lessened significantly(P<0.01) and the sucrose intakeof preference test decreased obviously(P <0.01).On the other hand, compared with theliver-qi depression model group, the total score of open field test of rats in Shuyucapsule, Radix Stellaviae and fluoxetine treatment groups increasedsignificantly(P<0.01, P<0.001, P<0.05),and the sucrose intake of preference testincreased remarkably(P<0.05,P<0.05,P<0.01), and there was no statistical differencesin the three treatment groups(P>0.05). Radix Stellaviae can not significantly improvethe reduction of depression in rat sugar intake, but there are certain role in trends. The results of gene chips with41000known genes showed that the expression of theHtr1a, Htr5b, Htr2c, Htr3a in hippocampus is different in the five groups. The resultsof real-time quantitative PCR are Similar to gene chips, to verify the reliability of theresults of four target genes.Conclusion: The model rats with liver-qi depression can be prepared successfully byusing chronic mild stress (CMS)method, and they can be evaluated effectively bysucrose preference test and open field test.The differences in expression of the Htr1a,Htr5b, Htr2c, Htr3a in hippocampus may have close relationship with liver-qidepression,and probably is one of the centre pathogenesis of liver-qi depression.TheShuyu capsule and Radix Stellaviae can correct the changes, which may be one of themechanism to treat depression emotion.
