The aim of the quality control of Traditional Chinese Medicine and Chinese herbal medicine is to meet the need of modernization and internationalization of CTM. In recent years,"return to nature" is becoming increasingly popular in the world, traditional chinese medicine and natural medicine has attracted extensive attention by the international medical recently. The Chinese herbal medicine market is very hot. The industry of Chinese traditional medicine is facing new chance for development as well as tough challenges. There exists substandard products in the market sometimes. To ensure the quality of traditional Chinese medicine is safety, effective, stable, and controllable. Firstly, we should ensure the sources of Chinese herbal medicines reliability, secondly, we should build up a unified standard of quality control of traditional Chinese medicine. Chinese traditional medicine is a complex multicomponent system, the fingerprint of traditional chinese medicine as an effective method to control the quality of TCM has become internationally recognized. Application of fingerprint technology in quality control of traditional Chinese medicine is a hot issue in our country, and also a development direction in the future. The implementation and application of the standardized and specified fingerprint of traditional Chinese medicine will greatly promote the modernization of Chinese medicine. But in order to achieve the standards of quality control of traditional Chinese medicine is not easy, it need a lot of practice. The main contents of the thesis include the following parts.Firstly, A method based on High performance liquid chromatography with photodiode array detector (HPLC-DAD) was developed for chemical fingerprinting analysis of Herba Ephedrae. The index of chromatographic fingerprint’s information content was utilized to optimize the fingerprint detection conditions, which reduced the time of analysis and increased the veracity of analysis greatly. The methodology study show that the method of HPLC for the fingerprint analysis was precise, valid and satisfactory. Meanwhile, heuristic evolving latent projection (HELP) method was employed to resolve the overlapped chromatographic peaks, this method greatly enhanced the accuracy of the qualitative and quantitative results. With the resolved pure spectra identified three compounds tentatively. Bioactive compounds were discussed in detail. The aforementioned methods giving great help to analyze the chemical compositions of Traditional Chinese Medicine and provide a basis for using the Traditional Chinese Medicine in right way.Secondly, chemical pattern recognition methods, such as similarity analysis (SA) and hierarchical clustering analysis (HCA) was utilized to analysis the fingerprints of the Herba Ephedrae samples from different origins of China. The result of the similarity analysis indicated that the majority fingerprints of the samples have good correlation with the standard fingerprint. Only a few samples which correlation is not very good was identified as doubtful samples tentatively. Then, the hierarchical clustering analysis was utilized to discriminate the differences of these samples. The results obtained demonstrated that most samples are kept in the similar clustering state comparison to the similarity analysis. However, using the HCA method more outlier was selected out than the similarity analysis methods. It appears that HCA exhibits stronger ability to display the chemical characters of samples. Furthermore, five marker constituents were firstly screened out to be the main chemical markers, which importantly contribute to classification Herba Ephedrae samples. This investigation shows that the developed methodology can be generalized in the research of quality control of herb medicines.The significance of this paper lies in established the fingerprint of Herba Ephedra, which was utilized to control the quality of Herba Ephedra, analysis the fingerprints of the Herba Ephedrae samples from different origins of China. This study provided an important basis for reasonably utilizing and protecting the reserves and for planting them in standard operating procedure. The conditions for detection and extraction provided some references for pharmaceutical companies to ensure the stability of the proprietary production process.
