The chromatographic fingerprint analysis is the trend of Chinese medicine quality control. The fingerprints would be affected by the chromatographic operating conditions and data processing. How to building a good chromatographic fingerprint has been the focused and difficult problem of the fingerprint research.The purpose of this paper was trying to build an accurate and comprehensive chromatographic fingerprints so as to provide some references for Semen Armeniacae Amarae and similar medicines. Systematic work of the Semen ArmeniacaeAmarae chromatographic fingerprints has not been reported in the literature.The pretreated samples often contented a main component with high content and low levels of other minor components. It was a traditional method to construct the fingerprint at the maximum UV absorption wavelength of the active ingredient, which deviated from the integrity of fingerprinting. The full wavelength chromatographic fingerprint developed by us could display all the components in the fingerprint, which meets the integrity requirement of fingerprints well.Semen Armeniaca Amaraes from Gansu, Heilongjiang, Jilin, Hebei, Shanxi etc. with13batches of authentic ingredients, and28markets across the country were collected. It was the first time to create the fingerprint of Semen ArmeniacaeAmarae by HPLC. By the concept of total ion chromatography in gas-mass spectrometry, a full wavelength chromatographic fingerprints by adding UV absorption from210nm to360nm wavelength were constituted, which had better stability and reliability.The extract separation conditions have a significant impact on the content of amygdalin. In order to improve the extraction rate of amygdalin, the impact of different extraction solvents and methods were studied. The method of single factor was used to optimizate the extraction and separation of amygdalin.Methods such as principal component analysis, similarity calculations and cluster were used to analysis origin distinction about41batches of medicinals. The result was consistent with the origin of climatic conditions on herbs. In the principal component analysis, four kinds of active substances were discovered. Amygdalin was analysised in a qualitative and quantitative, and the other three kinds of active compounds was analysised in semi-qualitative.This work distinguished the difference between the different origins of Semen Armeniaca Amaraes, and provided a reference for pharmaceutical companies to purchase authentic original ingredients. The separation and extraction conditions provided some references for pharmaceutical companies to ensure the stability of the proprietary production process. The research of amygdalin active material would help structure-activity research and comprehensive development of Semen Armeniaca Amaraes. The total wavelength of fingerprint was put forward, which would provided a new idea for fingerprint technology research.
