ObjectiveTo investigate the effect of heat shock protein70(HSP70) antisense oligodeoxynucleotides(ASODN) on proliferation, apoptosis and chemosensitivity of HeLa cells induced by cisplatin.Methods1. Hela cell line were transfected by antisense oligodeoxy-nucleotides, or sense> random oligonucleotides.There were four groups in this experiment which was control, S,AS and R group. The expression of HSP70was detected by western blot.2. Hela cell line were induced by cisplatin. There were four groups in this experiment which was control, Cis, AS+Cis, S+Cis, R+Cis group. The innibitory rates on proliferation cervical cancer Hela cells treaded by cisplatin was determined using methyl thiazolyl tetrazolium(MTT). The apoptosis rate was detected by flow cytometry.Results1. Intensity ratio of HSP70in each group(control5S,AS and R group) were (1.37±0.19),(0.38±0.13),(1.40±0.16)>(1.41±0.16), The protein expression of HSP70in Hela cells was significantly downregulated by HSP70ASODN(P<0.01). The results indicated ASODN can effectively inhibited expression of HSP70of cervical cancer Hela cells. 2. The inhibitory rates on proliferation of Hela cells in each group(control, Cis, AS+Cis, S+Cis, R+Cis group) were (100.00±6.71)%、(81.29±5.42)%、(61.79±3.18)%、(81.72±6.41)%(80.00±7.12)%. The inhibitory rates on proliferation of Hela cells in AS+Cis group was significantly higher compared with those of other groups (P<0.01). The results indicated inhibiting expression of HSP70can enhance cisplatin on Hela cells growth inhibition.3. The apoptosis rate of Hela cells in each group(control, Cis, AS+Cis, S+Cis, R+Cis group) were (4.32±1.57)%、(14.86±2.65)%、(24.61±2.90)%、(16.63±3.01)和(16.01±2.71)%. The apoptosis rate of Hela cells in AS+Cis group was significantly higher compared with control (P<0.01) and Cis group (P<0.05). The results indicated inhibiting expression of HSP70can increase apoptosis induced by cisplatin.ConclusionHSP70ASODN can effectively inhibit the proliferation and promote the apoptosis of HeLa cells by inhibiting expression of HSP70. HSP70ASODN and cisplatin have synergistic effect, consequently improve tumor chemosensitivity. HSP70may thus become a new target of gene therapy for cervical cancer.
