Object:①To explore the expression of Her-2and the breast cancer stem cell marker ALDH1in breast cancer tissues and their correlationship.②To explore the role of Her-2and the breast cancer stem cell marker ALDH1for the value of survival analysis and prognosis of breast cancer.Method:The breast cancer tissues were collected from105cases who accepted the eradicative operation from2008to2010and the protein expressions of the special genetic markers, her2and ALDHl were detected using immunohistochemical method detection. Combined with the clinic pathological correlation analysis and disease-free surial analysis, the independent risk factors for the prognosis of patients were evaluated.Results:①The positive expression rates of Her-2and ALDH1in breast cancer were29.5%and66.7%respectively, and the positive expression rates of Her-2and ALDH1were37.5%and75%in48patients with axillary lymph node metastases.②The protein expression of Her-2in breast cancer was positively correlated with tumor size an negatively correlated with ER (P<0.05), and there are no correlationship between the protein expression of Her-2and age, nationality, histological staging, pathological type and PR(P>0.05). There are no correlationship between the protein expression of ALDH1and age, ethnic, histological staging and pathological types, tumor size, lymph node metastasis, ER and PR (P>0.05).③The protein expressions of her2and ALDH1, the Special genetic markers in breast cancer stem cells were positively correlated(r=0.192, P<0.05).④In the continuous slice samples, the high expression region of her2and ALDH1were highly overlaped, the overlap rate is72%.⑤2years of disease-free survival rate was significantly lower in patients with high expression of Her-2than that in patients with negative expression of Her-2(P<0.05), the disease-free survival rates in patients with negative expression of Her-2and positive expression of Her-2were95.9%and77.4%respectively; there are no significant differences in2years of disease-free survival rates between patients with negative expression of ALDH1and positive expression of ALDH1(P>0.05), and the disease-free survival rates in patients with negative expression of ALDH1and positive expression of ALDH1were88.6%and91.4%, respectively.⑥Using COX prognosis of the multi-factor analysis model, the results suggest that Her-2is an independent prognostic factor (P=0.008). The relative risk of Her-2was6.284. indicating that the prognosis is lower in patients with positive expression of Her-2than that in patients with negative expression of Her-2. There are no significant diffrences between patients with negative expression of ALDH1and positive expression of ALDH1.Conclusion:①The protein expression of Her-2and ALDH1were positively correlated.②The protein expression of Her-2in breast cancer tissues was positively correlated with tumor size, and negatively correlated with ER.③the disease free survival rate of the breast cancer patients was decreased when the Her-2protein positively expressed. The results provide a certain basis that the Her-2and ALDH1may be used as a breast cancer prognostic indicators.
