Objective: To investigate the expression of angiopoietin-1,2mRNA in humanoral squamous cell carcinoma(OSCC)and nomal oral mucosa, and evaluate thedifference of expression in angiopoietin-1,2mRNA and protein in human oralsquamous cell carcinoma(OSCC)and nomal oral mucosa,and evaluate thecorrelation of angiopoietin-1,2mRNA and protein to clinicopathologicfeatures,tumor angiogenesis of OSCC. Methods: Collection of the SichuanProvincial Tumor Hospital Head and Neck Surgery to remove the submission ofspecimens of50cases.35cases of OSCC specimens, l5cases of normal oralmucosa. All specimens were confirmed by pathology. The expression ofAng-1/Ang-2protein and mRNA by Immunohistochemistry and semi-quantitative RT-PCR.CD34in tissue samples was detected by immunohistochemicalstaining.Microvessel density(MVD)in tissue samples was counted. Results:Ang-2mRNA was detected in OSCC and noncarcinomaous oral tissues.Thepositive rate of Ang-2mRNA was significantly higher in OSCC than in nomaloral mucosa(P<0.01).MVD was positively correlated with the expressiondegree of Ang-2mRNA. The expression of Ang-2mRNA was significantlyhigher in muscle invasive OSCC(P<0.05).No significant difference wasfound between the expression of Ang-1mRNA in nomal oral mucosa andnomal oral mucosa(P>0.05). Ang-1protein in oral squamous cell carcinomawas significantly lowered. Ang-2protein was significantly up-regulated. Thereare Significant difference(P<0.05). Ang-2positive expression of OSCC meanMVD48.10. Negative expression of OSCC mean MVD38.14. The latter wassignificantly lower than the former(t=6.871,P<0.05). Microvessel density inoral squamous cell carcinoma and oral squamous cell carcinoma patient age,gender, histological grade, clinical stage, lymph node metastasis was not statistically significant. Conclusions: Ang-1protein in normal oral mucosa ishigher than oral squamous cell carcinoma; Ang-2protein in oral squamous cellcarcinoma is higher than normal oral mucosa; Ang-2mRNA in oral squamouscell carcinoma is higher than normal oral mucosa; Expression intensity ofAng-2had correlation with invasion depth of oral squamous cell carcinoma,may play an important role in the malignant progression. Expression Ang-2hadcorrelation with more microvessel density of oral squamous cell carcinoma.Angiopoietin-2may take part in angiogenesis in oral squamous cellcarcinoma.The microvessel density of oral squamous cell carcinoma had nocorrelation with lymph node metastasis and clinical stage. That need to beconfirmed in a large series of OSCC patients.
