Background: Ion channels widely distributed with the organization organs,maintaining the normal physiological function and homeostasis in positiveimportant effect, IKCa1channel activation can lead to cell membrane atlong-term super polarization state, and promote the cell calcium ions in the flow,thus further induce complex cell signaling, which causes the cell proliferationand cause a variety of out of control the happening of the disease, even theformation of the tumor. In addition, in the human prostate cancer and pancreaticcancer, breast, colon, endometrial cancer tumor tissues or cells are found inIKCa1channel in high expression [1,2]. In spite of the current to thepathogenesis of cervical cancer have a certain understanding of, however, therelevant IKCa1in cervical cancer pathogenesis is still not clear.Objective: this study of cervical cancer cell lines through HELA IKCa1ion channels of expression, and observation of the tumor IKCa1block after theinfluence of cell proliferation cycle, this paper discusses IKCa1expression andcervical cancer of the potential contact and IKCa1happened in cervical cancerpossible mechanism. Methods: cervical cancer HELA cell lines as the researchobject, the construction of the IKCa1contains specific iRNA fragments ofpGenesil-IK1plasmid. Joint use fluorescent quantitative PCR, the WesternBlotting and molecular biology method and patch clamp electrophysiologytechnology, more pGenesil-IK1plasmid ZhuanRan or use IKCa1channel inhibitors intervention HELA cells before and after cerical cancer IKCa1genesand protein expression level and IKCa1current changes, and observe ZhuanRaninterference after plasmid HELA cell proliferation of influence. Results:(1)cerical cancer cells IKCa1current obvious enhancement, and can be itsspecificity blockers inhibited. PGenesil-IK1interference plasmid ZhuanRanHELA cells after current weakened obviously (P<0.05);(2) ZhuanRanpGenesil-IK1interference plasmid HELA cells after IKCa1mRNA and proteinexpression level is reduced the control group (P<0.05),(3) ZhuanRanpGenesil-IK1interference plasmid HELA cells of the growth period ofextended; Conclusion:(1) cerical cancer cells have IKCa1HELA onexpression, and calcium ion is of the dependent;(2) and normal cervical thanepithelial cells, cervical cancer cells IKCa1HELA on the expression of asignificantly higher level;(3) cerical cancer cell lines IKCa1HeLa inhibitiongene and protein expression, can effectively restrain cancer cell proliferation.⑷HELA cell membrane potential resting with the inner cell calciumconcentration and reduce increased;(5) HELA cell membrane potential can beIKCa1specificity blockers Clotrimazole and plasmid pGenesil-IK suppression,tip IKCa1can directly control HELA cell membrane potential thus influence itssignal transmission, cell division and proliferation.
