BackgroundCholangiocarcinoma is a malignant tumor of the biliary system, which diagnosis is difficult and has unknown etiology as well as poor prognosis. The number of cases has showed a growing trend in recent years, most of them were late detected and with high mortality rate. Lactate dehydrogenase (LDH) is one of the key enzymes of glycolysis. The LDH is a tetramer composed by four polypeptide chains, which encoded by two different genes. According to different ratio, it has five isoforms (LDH-1~LDH-5). The LDH-5(M4) is encoded by LDH-Agene, also known as LDHA, whose main function is to catalyze the conversion of the pyruvate into lactic acid. Studies have shown that tumor growth is closely related to LDH. There are different phenomena, such as serum LDH activity increasing and LDHA expression increasing in tumor cells. Owing to the cholangiocarcinoma has been study few in this field, this research detect the expression of LDHA in human cholangiocarcinoma tissues and cells.ObjectiveObserve the expression of Lactate dehydrogenase-A (LDH-A) in cholangio-carcinoma tissues, human cholangiocarcinoma cell lines HuCCTl and human intrahepatic bile duct epithelium cell lines HIBEpiC.MethodsThe expression of LDHA in32cholangiocarcinoma and15adjacent non- cancerous tissues was analyzed by Immunohistochemistry. RT-PCR and Western blot were used to detect the expression of LDH-A in HuCCT1and HIBEpiC.ResultsThe high expression of LDHA in cholangiocarcinoma tissues is18/32(56.3%, P<0.01) and also showed in HuCCT1cells but HIBEpiC cells performed as low expression.ConclusionCholangiocarcinoma tissues and human cholangiocarcinoma cell lines HuCCTl are both found LDHA high expression which indicate that LDHA may be an important role in initiation and progress of cholangiocarcinoma.
