| Objective To observe the dynamic changes of the proportions of T cytotoxic type1(Tc1) cells and T cytotoxic type2(Tc2) cells and analyze the correlation of themwith the dynamic changes of the proportions of Th1and Th2cells in T lymphocytesrespectively at different stages after Schistosoma japonicum infection. MethodSplenocytes were prepared from spleens of mice with S. japonicum infection atdifferent stages (0w,3w,5w,8w,13w), and the proportions of Tc1, Tc2and Th1,Th2cells in T cells were determined by FACS respectively. Spleen lymphocyteswere stimulated with SWA and SEA for48h in vitro, and the proportions of Tc1,Tc2and Th1, Th2cells in T cells were determined by FACS respectively. ResultsCompared to the control group (0w), the proportions of Tc1cells significantlyincreased3,5,8,13weeks post-infection (P<0.01, P<0.01, P<0.001, P<0.001) andthe proportions of Tc2cells at different stages (5w,8w,13w) post-infection alsoincreased significantly (P<0.01, P<0.01, P<0.001). During the first3weekspost-infection, the proportions of Tc1cells increased most quickly than Tc2cells didand Tc1cells did at the other stages post-infection. However, the proportion of Tc2cells increased most quickly during3-5weeks post-infection than both Tc1cells did and Tc2cells did at the other stages post-infection. A positive correlation was foundbetween the increases of the proportions of Th1cells and Tc1cells (r=0.978,P=0.004), and a positive correlation was also found between the increases of theproportions of Th2cells and Tc2cells (r=0.974, P=0.005). The SWA stimulationof splenocytes of mice in vitro significantly increased the proportion of Tc1cells(P<0.001) while the SEA stimulation of splenocytes of mice in vitro significantlyincreased the proportion of Tc2cells (P<0.001). Conclusion The proportions ofTc1cells and Tc2cells in T cells are significantly increased at different stages afterS. japonicum infection. Tc1cells are increased most quickly during first3weekspost-infection, and Tc2cells are increased most quickly during3-5weekspost-infection. A positive correlation is found between the proportions of Th cellsand Tc cells in CD3~+T cells. According to the rate of growth, Tc1and Tc2maypromote Th1and Th2differentiation respectively. SWA preferentially induces theincrease of Tc1cells in CD3~+T cells while SEA preferentially induces the increaseof Tc2cells in CD3~+T cells in vitro. |
