| Objective: We Isolated the stem-like cells in SW982cell lines, a synovial sarcomacell line；and took a Verification of its stem-like cell feature.Methods:①We culturedSW982cells in a low serum Medium, got the cells which can culture under the lowestconcentration. Then we had a Drug-resistant experimental.②We tested the inhibition rateof tumor cell cultured under different concentrations of adriamycin use MTT assay.③Inoculate both of drug-resistant and none drug-resistant cells with soft AGARmedium,observing the monoclone of cells；④W e tooka tumorigenicity experiments on thenude mice with the drug-resistant and none drug-resistant cells seperatively. Results:Drug-resistance capacity of SW982cell is improved after cultured with low concentrationof serum; In monoclonal experiment,the clonal volum of drug-resistant cells becomebigger,the number start to increase, growing in the a nest form three days afterinoculation,while none drug-resistance cells cannot live;In the tumorigenicity experiments,we found that there were two nude mice form a tumor after injected drug-resistant cells,the others did not form a tumor.Conclusion:①There are high tumorigenicity tumorstem-like cells existing in the SW982cell lines;②we can collect high tumorigenicitysynovial sarcoma cell line effectivlely with high concentration of adriamycin, but to gainsynovial sarcoma stem cells still need further purification. |
